RNA-seq is the next-generation sequencing based technique that we use to analyse cellular gene expression profiles. Depending on the purpose of the project, we regularly perform RNA-seq data to study differential gene expression, RNA editing events, or SNP identification.
Quality and quantity of the RNA sample is crucial for successful RNA sequencing, and we aim for the RIN (RNA Integrity Number) of 8 and above for samples extracted from frozen tissue or fresh cardiomyocytes to ensure quality data output. Our lab has also developed protocols to perform sequencing with low input of RNA, as low as 1ng.
